If you're starting from the Darwin city center, head northeast on Smith Street towards The Esplanade. Walk for about 10 minutes until you reach The Esplanade Park. Turn left onto The Esplanade and continue walking until you reach the intersection with Gilruth Avenue. Turn right onto Gilruth Avenue, and you'll find the entrance to the George Brown Darwin Botanic Gardens on your left after about 500 meters.
