If you're starting from the Agra Fort, exit the fort complex and walk towards the main road. Head southeast on Fort Road and continue walking for about 500 meters until you reach the intersection with Rakabganj Road. Turn right onto Rakabganj Road, and walk straight for another 300 meters. Diwan-e-Khas will be on your left at the entrance of the Agra Fort complex.

From your hotel or any central location in Agra, you can take an auto rickshaw to Agra Fort. Simply tell the driver 'Agra Fort' and they will take you there. The ride should take around 10-15 minutes, depending on traffic. Once you arrive at Agra Fort, exit the complex and follow the walking directions provided above to Diwan-e-Khas.
