If you're near the Faro Marina, exit the marina area and head towards Rua de 1º de Maio. Continue walking straight until you reach the intersection with Avenida da República. Cross Avenida da República and continue straight on Rua de Portugal until you reach Praça da Liberdade. Turn left onto Rua 1º de Maio, and continue walking straight until you arrive at Largo da Sé. The church will be directly in front of you.
Public Transport

You can take a local bus from the Faro Bus Station. Look for a bus that goes towards the city center, specifically one that stops near Largo da Sé. The stop closest to the church is called 'Sé'.
