From the Sistine Chapel, exit the Vatican Museums and proceed towards the entrance/exit gates. Once you exit, walk towards the main road (Viale Vaticano) and turn left. Continue straight until you reach Piazza San Pietro (St. Peter's Square). From there, you can take bus number 81 from the nearby bus stop (Piazza San Pietro) towards 'Piazza Venezia'.
Public Transport
Head to the nearest metro station, Ottaviano, which you can reach by walking straight from the Vatican Museums. Once at the Ottaviano station, take Line A (the red line) towards Anagnina. At the Termini station, transfer to Line B (the blue line) towards Laurentina. Get off at the 'Castro Pretorio' station and walk to the nearby bus stop to catch bus number 553 towards 'Frascati'.

Simply find a nearby taxi stand or request one through your hotel or an app. Tell the driver to take you to Tusculum in Frascati, and they will navigate the route for you, which typically takes about 40 minutes depending on traffic.
