Turn left onto the Chhukha Highway (also known as the Thimphu-Phuentsholing Road). Drive for about 7 kilometers, and then look for signs pointing to Semtokha. Keep an eye out for the turnoff towards Semtokha Dzong. Follow the signs for about 2 kilometers, and you will reach Simtokha Dzong, located at 846 Je Bamo Lam SE, Tshatshobaykha.
