From the Fresno Amtrak Station, walk to the nearest bus stop at Tulare St & G St. Take Bus Line 34 (Fresno State) towards the 'Fresno State' station. Get off at the stop at Fulton St & Tuolumne St. From there, walk north on Fulton St for about 0.3 miles, and the African American Museum will be on your left at 1857 Fulton St.
