If you are in the Harbourfront area, you can easily reach the Toronto Railway Museum by walking. Start at the Harbourfront Centre, located at 235 Queens Quay W. Continue walking along the waterfront until you reach the intersection of Queens Quay W and Lower Simcoe St. Turn left onto Lower Simcoe St and continue until you reach Bremner Blvd. The Toronto Railway Museum will be on your right at 255 Bremner Blvd. This walk should take approximately 15-20 minutes.
