From Hlane Royal National Park, head southeast on the main road towards the town of Big Bend. Continue on this road until you reach the town of Mhlume. At Mhlume, follow the signs directing you towards Mlilwane Game Sanctuary. The sanctuary is located approximately 30 kilometers from Mhlume. Follow the signs to the entrance gate where you can park your car and explore the sanctuary.

From Hlane Royal National Park, you can take a local minibus taxi towards the town of Big Bend. Once in Big Bend, transfer to another minibus taxi heading towards Mhlume. In Mhlume, you can either hire a local taxi to take you directly to Mlilwane Game Sanctuary or look for shared taxis heading in that direction. The sanctuary is approximately 30 kilometers from Mhlume. Ask the driver to drop you off at the entrance gate where you can start your visit.
