From Ein Gedi, walk to the bus stop located on Route 90. Board a bus heading north towards Jerusalem, which should take about 1.5 hours. Once you arrive in Jerusalem, disembark at the central bus station.

After reaching the central bus station in Jerusalem, take the light rail (tram) towards the 'City Hall' station. Get off at the 'City Hall' stop and walk approximately 15 minutes towards the Old City. Follow signs towards the Jaffa Gate, and from there, continue walking straight until you reach the Church of the Holy Sepulchre.

Alternatively, after reaching the central bus station in Jerusalem, you can take bus number 99 or 75 directly to the Old City. Get off at the 'Damascus Gate' and walk towards the Church of the Holy Sepulchre, which is around a 10-minute walk from there. Look for the signs leading to the church.
