If you're traveling by car from Mugello, start on the SS65 main road heading south towards Florence. Continue on SS65 for about 20 kilometers until you reach the outskirts of Florence. From there, follow the signs for 'Centro' to merge onto Viale Giovanni Amendola. After approximately 2 kilometers, turn left onto Via di Santa Caterina da Siena. This road will take you directly to Piazza San Marco. Note that parking in the area can be limited and may incur costs.
