If you're starting from the Buffalo Niagara Convention Center, head east on Franklin Street towards the intersection with East Mohawk Street. Turn left onto East Mohawk Street and continue walking until you reach Main Street. From there, turn right onto Main Street and walk south until you reach the intersection with NY-16 (also known as the Buffalo Skyway). Turn left onto NY-16 and proceed a few blocks until you see the Old Post Office on your right at 88-56 NY-16.
