If you are starting from the Main Market Square (Rynek Główny), head north towards the Cloth Hall (Sukiennice). Once you reach the Cloth Hall, exit the square and take a right onto ul. Floriańska. Continue walking straight for about 5 minutes. You will see Lost Souls Alley located at Floriańska 6 on your left-hand side, just before you reach the Barbican.
