From Huelva, head to the Huelva bus station (Estación de Autobuses de Huelva). You can take a direct bus to Valencia, which usually takes around 8 to 10 hours. Make sure to check the bus schedules in advance. Once you arrive at Valencia's bus station (Estación de Autobuses de Valencia), exit the station and head to the nearby subway station.

At the Valencia bus station, take Line 3 or Line 5 of the MetroValencia towards 'Aeroport' or 'Rafalafena', depending on the line you choose. Travel for about 5 stops and get off at 'Xàtiva' station. From there, transfer to Line 1 (the yellow line) towards 'Bétera'. After one stop, get off at 'Colón' station.

Once you exit Colón station, head towards the 'Plaza de la Virgen'. Walk straight down Calle de la Paz until you reach Plaza de la Reina. From Plaza de la Reina, continue walking towards the 'Plaza de Manises'. The Tower of Saint Bartolome will be located at Pl. de Manises, 3A, Ciutat Vella, which is a short walk from the Plaza.
