If you are starting from the Ark Fortress, exit the fortress and head south towards the Registan. Continue straight onto Kosh Madrasah Street, and eventually, turn left onto Mehtar Ambar Street. Toqi Telpakfurushon will be located on the right at number 37. The walk should take about 15-20 minutes.
