Walking
From the entrance of the Sistine Chapel, exit the chapel and turn left onto Viale Vaticano. Continue walking straight until you reach the main road, Via di Porta Angelica. Cross the street and continue straight until you reach the intersection with Via Della Conciliazione. Turn right onto Via Della Conciliazione and walk until you see St. Peter's Basilica on your left. From there, you will continue straight until you reach the end of the street, then turn left onto Via della Conciliazione. Keep walking for about 1 km until you reach the entrance of the Vatican Museums. The Roma Experience Tours office is located at Via Oderisi da Gubbio, 13A, which is a 15-20 minute walk from the Vatican Museums.

Public Transport
From the Sistine Chapel, walk back to the Vatican Museums. Once there, head to the nearest bus stop, which is located on Viale Vaticano. Take bus number 49 in the direction of 'Cinecittà'. Stay on the bus for about 6 stops and get off at the stop called 'Piazza di Porta Maggiore'. From there, you will need to transfer to tram line 14. Board the tram towards 'Porte di Roma'. After about 10 minutes, get off at 'Piazza dei Cinquecento'. Walk 10 minutes to Via Oderisi da Gubbio, 13A. The entire journey should take approximately 35-45 minutes.
