Walking
From the Sistine Chapel, exit through the main entrance to the Vatican Museums. Once outside, turn left and walk towards the entrance of St. Peter's Square. Continue walking straight, and you will see the large dome of St. Peter's Basilica ahead. After passing St. Peter's Basilica, proceed straight, following the street that leads you out of the Vatican area. After about a 10-minute walk, you will reach Via della Conciliazione. Cross the street and continue walking straight until you reach the intersection with Via dei Cestari. Turn right onto Via dei Cestari and continue until you reach Piazza Mattei, which will be on your left. The entire walk should take approximately 25-30 minutes.

From the Sistine Chapel, exit through the main entrance of the Vatican Museums. Walk towards the bus stop located at 'Piazza San Giovanni in Laterano'. You may take bus number 64, which travels towards 'Termini'. After a few stops, get off at 'Piazza Venezia'. From there, you can either walk to Piazza Mattei, which is about a 10-minute walk away, or you can catch bus number 87 from the same bus stop to 'Piazza Mattei' directly. The total travel time using public transport should be around 30-40 minutes, including walking.
