If you're starting from downtown Denver, head to the Union Station light rail station. Take the E Line towards the Denver Convention Center. Ride for about 6 stops and disembark at the 'Colfax at Auraria' station. From there, transfer to the 15 bus line heading east towards 'York St & 10th Ave.' Get off at the 'York St & 10th Ave.' stop. The Denver Botanic Gardens will be a short walk from there, just two blocks north.
