If you're starting from the Old Town of Rhodes, exit through one of the gates and head towards the waterfront. Walk along the coast toward the east for about 20 minutes until you reach the Mandraki Harbor. You'll see the iconic Colossus of Rhodes statue. Continue walking along the promenade. The Saint Nicholas Fortress will be visible at the end of the harbor on a small island. You can walk to the fortress by following the path that leads to it.

From the main bus station in Rhodes Town, take a bus heading towards Lindos or any bus that stops at the Mandraki Harbor. The ride should take about 10 to 15 minutes.
