If you are starting from Bhopal, head east on NH46 towards Hoshangabad. After approximately 60 km, exit onto the road towards Ratibandar (look for signs indicating Bori Wildlife Sanctuary). Continue straight for about 20 km, and you will arrive at the sanctuary entrance. Ensure you check the condition of the roads, especially during the monsoon season, as some routes may be affected.
