If you are starting from the Montepulciano train station, exit the station and head north towards the main road (Via di Gracciano nel Corso). Once you reach the main road, turn right and continue walking straight. Follow the road uphill for about 2 km until you reach the town's entrance. Once you enter Montepulciano, continue along the main street (Corso di Vannucci) which will lead you through the town. As you walk, you will pass several shops and cafes.
