Start at the main square, Piazza IV Novembre, which is the heart of Perugia. From the square, head south towards Corso Vannucci, the main shopping street. Continue down Corso Vannucci until you reach the intersection with Via Mazzini. Turn left onto Via Mazzini and continue straight until you reach the end of the street where it meets Via Appia. Turn right onto Via Appia, and after a short walk, you will find Scalinata dell'Acquedotto on your left.
