If you are starting from the Pompeii Archaeological Park entrance, head straight down Via Plinio towards the main road, Corso Giuseppe Garibaldi. Turn right onto Corso Giuseppe Garibaldi and continue walking until you reach the intersection with Via Marina. Turn left onto Via Marina. Continue walking for about 500 meters, and you will find Casa di Romolo e Remo on your left at Via Marina, 2.
