If you are near Piazza San Marco, head northwest to the water's edge and take the Fondamenta dei Canonici path. Continue along this path until you reach the Ponte della Paglia. Cross the bridge and turn left onto Calle del Rimedio. Follow this street straight until you reach Campo Santa Maria del Giglio. The church will be on your right.

From the Vaporetto stop, walk straight into the campo (square) and the Chiesa di Santa Maria del Giglio will be right in front of you.
