If you prefer public transport, take the Tokyo Metro Toei Oedo Line from Roppongi Station to Akasaka-Mitsuke Station. It’s just one stop away. From Akasaka-Mitsuke Station, take Exit 1b and walk towards Roppongi Hills. Follow the signs to Mori Tower; it’s about a 15-minute walk from the station. Note that the metro fare is approximately 220 yen.

For a quicker option, you can take a taxi. Simply hail a taxi from anywhere in Roppongi and tell the driver 'Roppongi Hills Mori Tower'. The ride usually takes around 5 minutes, depending on traffic.
