If you're driving from Macina, head northwest on the main road, Route N6. Continue straight for about 220 km until you reach Bamako. As you approach the city, follow the signs for the city center. Once in Bamako, take Avenue Moussa Tavele and continue to M252. The National Museum is located at M252+C6C, close to the banks of the Niger River. Make sure to park in a safe area nearby.
