From the bus stop, you can take a local bus to Victoria (the capital city of Gozo). The bus will take approximately 20 minutes. Once you arrive in Victoria, disembark at the 'Victoria' bus terminal. From the terminal, walk towards Charity Street. The Basilica is located at 26VQ+HJM Charity Street, which is a short 5-minute walk from the terminal.
