If you are traveling by car, start from any point in the South Eastern District and head towards Valletta. Follow signs for 'Sliema' and 'Gżira' on the main road (Triq Sant' Anna). As you approach Gżira, look for the signs directing you to Manoel Island. You will need to find parking nearby as there is limited space on the island itself. The closest parking area is along Triq Manoel, which is a short walk to Fort Manoel.
