If you are starting from the Upper Barracca Gardens, exit the gardens and head down the stairs towards the Valletta Waterfront. Once you reach the bottom, turn left and walk along the waterfront promenade until you reach the main street. Turn right onto the main street and continue straight. You will pass through Republic Street, which is lined with shops and cafes. Keep walking until you reach St. Mark Street; the Fortifications Interpretation Centre will be on your right.
