If you are driving from Windhoek, take the B1 highway south towards Rehoboth. Continue on the B1 until you reach the turnoff to the C24 road towards Sossusvlei. After paying the fee, continue on the road to Sossusvlei. Once you arrive at the Sossusvlei parking area, you will need to take a 4x4 shuttle (costs around 100 NAD per person) to reach Deadvlei, as the last part of the road is not accessible by standard vehicles.
