From the center of Leiden, head towards the train station (Leiden Centraal). Exit the station and walk to the bus stop in front of the station. Take Bus 43 towards The Hague (Den Haag) and get off at the 'Den Haag, Plein' stop. From there, it is a short walk to Mauritshuis. Head towards Plein 29; the museum will be directly in front of you.

From there, you can take tram line 9 towards Scheveningen and get off at 'Plein.' Walk straight to Plein 29 where Mauritshuis is located.
