If you're driving from any location in The Hamptons, take NY-27 W (Sunrise Highway) towards Riverhead. Continue on NY-27 W for about 25 miles until you reach the exit for NY-24 N towards Riverhead. Merge onto NY-24 N and continue for about 5 miles until you reach the intersection with E Main St. Turn right onto E Main St, and after about 1 mile, you will see Long Island Aquarium on your left at 431 E Main St, Riverhead, NY 11901.
