From Lehigh Valley, take I-78 E towards Allentown. Merge onto I-476 S (the Pennsylvania Turnpike) and continue for about 20 miles. Take exit 16A to merge onto I-76 E toward Philadelphia. Stay on I-76 E for approximately 20 miles. Take exit 344B toward US-1 N. Merge onto US-1 N and continue for about 1 mile. Use the right 2 lanes to merge onto I-95 S. Take exit 22 toward Callowhill St, then keep right at the fork and follow signs for Fairmount Ave. Continue onto Fairmount Ave, and you will find Eastern State Penitentiary at 2027 Fairmount Ave, Philadelphia, PA 19130. Parking is available nearby but may incur a fee.

From Lehigh Valley, you can take a SEPTA Regional Rail train from the Allentown station to Philadelphia's 69th Street Terminal. Once in Philadelphia, transfer to the Market-Frankford Line (blue line) and travel to 69th Street. From there, take the Route 7 bus towards the Art Museum; the bus stops at Fairmount Avenue, a short walk from Eastern State Penitentiary located at 2027 Fairmount Ave.
