From the Little India MRT Station, head east on Serangoon Road. Continue straight for about 600 meters until you reach the intersection with Jalan Besar. Turn right onto Jalan Trus and follow the road for about 1.2 kilometers until you reach Lot 653, where the Johor Ancient Temple is located.
