From the center of Cuenca, head to the main bus terminal (Estación de Autobuses de Cuenca). Look for the bus lines that go to Biblián. Take the bus towards Biblián, which should take approximately 30 minutes. Once you arrive at Biblián, get off at the stop closest to Av. Daniel Muñoz. From there, walk straight along Av. Daniel Muñoz for about 10 minutes, and you will reach the Sanctuary of the Virgen del Rocio.

If you prefer a more direct route, you can take a taxi from Cuenca to the Sanctuary of the Virgen del Rocio. Simply flag a taxi or use a ride-hailing app. Inform the driver that you want to go to Av. Daniel Muñoz in Biblián. The journey should take around 20-25 minutes, depending on traffic.
