If you are starting from the Valencia Cathedral, head southeast on Plaza de la Reina towards Calle del Mar. Continue straight until you reach the intersection with Calle de la Paz. Turn left onto Calle de la Paz and walk until you reach the next intersection. Turn right onto Plaza de la Virgen. The Basilica de la Mare de Déu dels Desemparats is located at the far end of the square, at Plaça de la Verge, s/n.

Take Line 3 or Line 5 from any metro station in Valencia that connects to the metro system (e.g., Xàtiva or Colón). Get off at the 'Colon' station and transfer to Line 3, heading towards Rafelbuñol. Get off at the 'Àngel Guimerà' station. Exit the station and head southeast on Avenida de la Paz until you reach the intersection with Plaza de la Virgen. The Basilica will be on your right.

Take bus line 95 or 99 from any nearby bus stop. Make sure to check the direction and get off at the stop 'Plaza de la Virgen'. From there, you can walk directly to the Basilica de la Mare de Déu dels Desemparats, which is located at the end of the square.
