From the Matara Bus Station, head east on Matara Rd towards Galle Rd. Continue straight for about 1 km until you reach the junction with Galle Rd. Turn right onto Galle Rd and continue walking for approximately 1.5 km until you reach the entrance to Galle's city area. Once you enter Galle, follow the signs towards Church St, which is located in the heart of the city. You will find Church St on your left after a few minutes of walking.
