Walking
If you are at Wat Mahathat, start by walking south on Naresuan Rd for about 500 meters. Turn right onto U-Thong Rd and continue walking north for another 1 km. Watch for signs directing you to Wat Tawed, which will be to your right. The entrance is a short walk from the main road.
Tuk-Tuk
From your current location in Ayutthaya, hire a tuk-tuk and inform the driver that you want to go to Wat Tawed. The fare should be around 60-100 THB depending on your starting point.
