Walking
Exit the Gregorian Etruscan Museum and head towards the main entrance of the Vatican Museums. Once you reach the main entrance, turn right and walk towards the entrance of the Vatican City. Continue straight down the Via di Porta Angelica until you reach the intersection with Via del Pellegrino. Turn left onto Via del Pellegrino and walk straight until you see the sign for the Big Bus Tours Rome on your right. The bus stop is located near the corner of Via del Pellegrino.

From the Gregorian Etruscan Museum, walk to the nearest bus stop located on Via di Porta Angelica, just outside the Vatican Museums. Board the bus number 81 in the direction of 'Stazione Termini'. After about 10 minutes, get off at the 'Piazza San Giovanni in Laterano' stop. From there, walk towards 'Via delle Terme di Caracalla' and look for the Big Bus Tours Rome office, which is around a 5-minute walk from the bus stop.
