Walking
Exit the Sistine Chapel and head towards St. Peter's Basilica. As you exit, you will find yourself in the Vatican Museums courtyard. Walk straight ahead towards the main entrance of St. Peter's Basilica. Once you reach the entrance of St. Peter's Basilica, continue walking along the Via della Conciliazione, which leads out of Vatican City towards Rome. Follow this road until you reach the intersection with Via di San Giovanni in Laterano. Turn right onto Via di San Giovanni in Laterano and continue walking for approximately 10 minutes. You will cross several streets; keep an eye out for signs indicating the direction to Palazzo Mattei. After walking for about 800 meters, you will arrive at Palazzo Mattei, located on Via Michelangelo Caetani, 32.

From the Sistine Chapel, exit and turn left towards the Vatican Museums exit. Walk through the courtyard and exit the Vatican Museums. Once outside, turn right and head towards the large street, Via della Conciliazione. Cross the street and continue straight ahead until you reach the Tiber River. Turn left and walk along the riverbank. Follow the river for about 1 kilometer until you reach the Ponte Fabricio. Cross the bridge and continue straight. You will find yourself on Via di San Lorenzo in Lucina, which will lead you directly to Palazzo Mattei after approximately 15 minutes of walking.

After visiting the Sistine Chapel, exit and go towards the Vatican Museums entrance. Once outside, head west towards the main road, Via della Conciliazione. Walk straight along this road for a few blocks until you reach the intersection with Via del Banco di Santo Spirito. Turn left onto this street and continue walking until you reach the intersection with Via Michelangelo Caetani. Turn right on Via Michelangelo Caetani, and you will find Palazzo Mattei at number 32 on your left side after a short walk.
