Walking
If you are starting from the Markt square, head southwest towards the Burg square. Continue walking straight until you reach the Burg square. From the Burg, take the road that leads to the left of the Basilica of the Holy Blood, which is called Wollestraat. Walk straight along Wollestraat until you reach the canal. Once you reach the canal, turn right onto Dijver. Continue walking along Dijver, and you will find the Gruuthusemuseum at number 17 on your left. The museum is located right by the canal, making it a beautiful walk.

If you are starting from the train station, exit the station and turn left onto Zilverstraat. Walk straight until you reach the main street, which is called Stationsplein. Cross the street and continue straight onto Langestraat. Follow Langestraat as it turns into Wijngaardstraat, then continue straight until you reach 't Zand square. From 't Zand, head towards the Markt square. Once at the Markt, follow the previous directions from the Markt to the Gruuthusemuseum.
