From Wilmington, take I-95 South to DE-1 South towards Dover. Take the exit for DE-896 South toward Middletown. Continue on DE-896 South until the intersection with US-13 South. Turn right onto US-13 South and follow it until you see signs for Port Penn. Turn left onto W Market St. The Port Penn Interpretive Center will be on your right. Parking is available nearby. There are no tolls on this route.

From Wilmington, take the DART Bus Route 40 to the New Castle Transit Center. Transfer to Bus Route 63 towards Port Penn. Get off at the Port Penn stop and walk about 0.5 miles to the center on W Market St. A one-way fare on DART First State buses is $2.00.
