From the Big Cypress National Preserve, head northeast on US-41 E towards Immokalee. Continue for about 20 miles until you reach the intersection with SR-29. Turn left (north) onto SR-29. After approximately 5 miles, you will see the entrance to the Florida Panther National Wildlife Refuge on your right at 12085 SR 29 South, Immokalee, FL 34142.
