Marshrutkas depart from the bus stop near Tbilisi's railway station, with the last departure typically around 4 PM (5-6 PM in summer). The ride to Manglisi takes approximately 1.5 hours and costs around 6 GEL. From Manglisi, a 20-minute walk will bring you to the park's entrance. Be prepared for a small fee of 1-2 GEL for the marshrutka.
