From the Mtskheta town center, head towards the main street, which is located near the Mtskheta Cathedral. Walk straight along the street (Aghmashenebeli Avenue) for about 500 meters until you reach the intersection with the main highway (E117). At this intersection, you will see a small park on your right side. Turn right and continue walking along the side of the park until you reach the bus stop in front of the park. Here, look for a public minibus (marshrutka) heading to Tbilisi.
