From Uplistsikhe, head west on the main road (E60) towards Gori. After approximately 10 kilometers, you will see signs for Gori. Continue straight until you reach the city center. Once in Gori, follow the signs to Gori Fortress, which is located at 10 Besarion Jughashvili St. There is parking available near the fortress, but be prepared to pay a small fee.
