Walking
If you are in the Friedrichshain area, the Berlin Wall site at Mühlenstraße 4-5 is easily accessible on foot. From the popular Boxhagener Platz, head southeast on Boxhagener Straße toward the main road, which is Karl-Marx-Allee. Continue walking straight until you reach the intersection with Mühlenstraße. Turn right on Mühlenstraße, and you will see the Berlin Wall memorial and the East Side Gallery just a short distance ahead. It's approximately a 20-minute walk.
Public Transport - Tram
You can also take public transport to reach the Berlin Wall. From Frankfurter Tor (U-Bahn station), take Tram M10 towards Warschauer Straße. Get off at the 'Schlesisches Tor' stop. From there, walk along the Spree River towards the East Side Gallery, which will take about 10 minutes. The tram ride costs around €3.00 for a single ticket.
Public Transport - Bus
Another option is to take Bus 140. From the nearest stop, 'Wühlischstraße,' board the bus heading towards 'Schönhauser Allee.' Get off at the 'Ostbahnhof' stop, and walk along the river towards Mühlenstraße. This walk will also take around 10-15 minutes and will give you a nice view of the area. The bus fare is the same as the tram, approximately €3.00 for a single ticket.
