If you are in Heraklion, take the KTEL bus from the Heraklion Bus Station to Rethymno. The journey takes about 1.5 hours. Once you arrive at the Rethymno bus station, exit the station and walk straight towards the main road (Leoforos Ethnikis Antistaseos). From there, turn left and walk towards the old town. Follow the signs directing you to the Fortezza, which is located about a 15-minute walk from the bus station.
