Start by heading to Route 44, which will connect you to Route 1 towards Jerusalem. Follow the signs for Jerusalem and then take the exit for the Old City. Once you are in the Old City, look for signs directing you to the Western Wall (Kotel). Note that parking can be limited in the Old City, so you may need to park in a designated area and walk to the Western Wall, which is about a 15-minute walk from most parking lots.
