If you are starting from the Florence Cathedral (Duomo), head southwest on Via dei Servi towards Piazza della Repubblica. Continue straight until you reach Piazza della Repubblica, then turn left onto Via degli Strozzi. Follow this street until you reach Via della Vigna Nuova, then turn right. Continue walking, and you will find Rucellai Palace at number 18 on your left.
Public Transport (Bus)
From Florence Santa Maria Novella Train Station, take Bus Line 11 towards 'Piazza della Libertà'. Get off at the 'Piazza della Repubblica' stop. From there, walk southwest along Via degli Strozzi, then turn right onto Via della Vigna Nuova. Rucellai Palace will be at number 18 on your left.
