If you are driving from the Tas-Silġ area, head south on Triq il-Knisja towards Marsaxlokk. Continue straight for about 2 kilometers until you reach the roundabout. Take the first exit onto Triq il-Lampuka, and follow the road until you see signs for Marsaxlokk Harbour. The harbour will be on your left. There is parking available nearby, but be aware that it may incur a small fee depending on the time of year and day.
